Fredericamycin A Derivatives
Fredericamycin A is a polyketide with antitumor activity derived from the soil bacterium Streptomyces griseus. It represents a new structural class of anti-tumor molecules. At present, about 250 derivatives of Fredericamycin A have been screened and a few compounds with potent, low nanomolar action and increased tumor selectivity have been identified. Their activity is comparable to the anti-tumor activity of cancer drugs like Doxorubicin and Topotecan.
Fredericamycin blocks several enzymes involved in DNA metabolism, e.g. DNA topoisomerase I and topoisomerase II as well as PIN1 (peptidyl-prolyl cis/trans isomerase), an enzyme involved in the regulation of the cell cycle and cell division.
